The modal MIC of GR69153 for the penicillin-susceptible strains was 0.12 , ug ml. Of the anaerobic bacteria tested, Bacteroides fragilis and Clostridium difficile were resistant MIC, 32 , ug ml; data for three strains not shown ; but other Clostridium spp. and anaerobic cocci were susceptible to GR69153 modal MIC, 0.25 pug ml; data not shown ; . In Table 3, the effects of serum on the MIC and MBC of GR69153 are shown. In the absence of serum, there was little or at most a two- to fourfold difference between the MIC and the MBC. The addition of 20 or 70%o serum had little consistent effect, the members of the family Enterobacteriaceae being somewhat more susceptible to GR69153 in the presence of serum and Staphylococcus aureus being less susceptible. The protein-binding activity of GR69153 was 74.5% at 5 , ug ml, 68.7% at 50 , ug ml, and 66.8% at 100 ug mnl. An increase in inoculum size from 10' to 106 CFU data not shown ; was accompanied, in general, by a fourfold increase in the MICs for gram-negative bacilli including P. aeruginosa ; and little or no change in the MICs for staphylococci. In Table 4, the relative efficiencies of hydrolysis of GR69153 and other agents in comparison with that of cephaloridine, which was 100% ; are shown. Meropenem and imipenem were highly resistant to hydrolysis by the range of enzymes studied. Amoxycillin was efficiently hydrolyzed by the TEM series, SHV-1, and K-1 enzymes. The TEM-3 enzyme hydrolyzed cefotaxime and had some minor effect on ceftazidime and GR69153. The TEM-9 enzyme displayed significant activity against all three cephalosporins and in particular GR69153. The chromosomal P99 enzyme did not hydrolyze imipenem or meropenem, but there was significant activity against GR69153. The efficiency of hydrolysis of GR69153 by TEM-9 and P99 was related to low Km values 9.8 and 4.4 , umol, respectively ; rather than higher Vmax values.
