Clinical response rates to MabCampath in refractory patients with B-CLL range from to 21% to 100%, with CRs of 0% to 80% see Table 1 ; . In the pivotal trial, CAM 211, 28 median time to progression was 4.7 months overall and 9.5 months for responders, while median survival was 16 months overall and 32 months for responders. This compares favourably with the median survival of 10 months seen in a similar cohort of 147 Fludara-refractory patients who received alternative, less effective treatment.29 MabCampath is highly effective in blood and bone marrow, 28, 30 and complete eradication of malignant cells can be observed at these sites of disease, even when novel, highly sensitive methods of detection are used, such as polymerase chain reaction or minimal residual disease-flow cytometry.31, 32 MabCampath can also dramatically reduce the bulk of affected lymph nodes, although complete disease eradication is best seen in lymph nodes of diameters less than 5 cm.28 The sites of disease at which MabCampath is most effective blood and bone marrow ; are those with the gravest consequences for the patient. Bone marrow infiltration causes anaemia and thrombocytopenia and increases susceptibility to infection. Infection is a major cause of death in patients with advanced B-CLL, 33 and the risk of serious infection increases markedly when patients have become refractory to Fludara therapy, with the incidence of hospitalisation increasing from 0.03 to 0.17 admissions per month.34 However, patients who respond to MabCampath and experience benefits in normal bone marrow function will also have improved immune function and a decreased risk of infection. Improvements in quality of life are evident even in patients who do not achieve clinical response; in the pivotal trial, in addition to the 33% of patients who responded, an additional 54% of patients achieved stable disease.28 Benefits included resolution of Bsymptoms night sweats, fever ; and fatigue, reduction of bulky lymph nodes and an increased chemotherapy-free period. A good response rate 40% overall response OR ; , 7.5% CR ; and a pronounced ability to stabilise disease and to confer quality-of-life benefits have also been reported in end-stage patients treated on a compassionate basis.35 Notably, MabCampath has also been shown to achieve responses in patients with p53 mutation deletion, a patient cohort that has a dismal prognosis when treated with chemotherapy.36 MabCampath, therefore, offers effective treatment for patients in whom current therapy options have reached their limits.

During the course of a study designed to characterize molecular lesions present in T-cell acute lymphoblastic leukemia T-ALL ; patients at diagnosis, we studied T-lymphoblasts from a T-ALL patient RUPN no. 78 ; obtained 16 hours after the institution of combination chemotherapy consisting of vincristine, doxorubicin, cyclophosphamide, and prednisone. Because most MLL gene rearrangements cluster within an 8.3-kb genomic DNA breakpoint cluster region MLL bcr ; , "~ * can be detected and by a 0.7-kb cDNA probe that encompasses this fragment, we screened genomic DNA extracted from patient lymphoblasts for MLL gene rearrangements by Southern blot using this 0.7-kb cDNA fragment. The results shown in Fig 1 initially suggested that an MLL rearrangement had OCcurred, with rearranged bands seen using four different restriction enzymes see arrows ; . However, the intensity of the rearranged bands compared with the germline bands was significantly lower than expected, and made us question this interpretation. Although white clover contribution to leaf density increased, its contribution to the total cover apparently did not increase in comparison to species. Further, according to total herbage, ground cover, and tiller leaf density other species did not change over the season. However, the other species contributed significantly more harvested herbage in autumn than they did in spring and summer. Since the proportion of white clover and other species in the harvested herbage and the percentages measured with the other methods changed with the seasons and were not consistently correlated with harvested herbage over the seasons, they would not be useful predictors of harvested herbage Fig. 3a, b ; . In addition, on average white clover contribution doubled from spring to summer, quackgrass percent contribution decreased by 4% in summer, and bluegrass percentage decreased by 8% in autumn Fig. 4 ; . The contribution of bluegrass may have decreased from spring and summer to fall because relative to other species, bluegrass growth decreased more than other species after the hot, dry summer conditions, and in comparison to the spring when tiller production and growth is usually high due to reproductive tiller development Mitchell, 1956; Wedin and Huff, 1996 ; . Bluegrass may also have allocated more reserves to belowground rhizomes in autumn. The contribution of quackgrass, a cool-season grass, decreased by 4% on average across methods in summer, probably due to the warm and drier summer conditions. Therefore, describing the botanical composition of a pasture on the basis of sampling once during one or even two seasons did not accurately represent the contribution of some species during spring, summer, and autumn and mechlorethamine.

The colony forming units correlated with the FACS data, dropping significantly for the vector alone and mph1 noninducing samples, increasing for the MPS1 sample and remaining the same for the mph1 sample data not shown ; . In addition, flow cytometric analysis of the DNA content of surviving mph1 containing cells was consistent with a partial rescue: while only 5 20 survivors in the non-induced sample remained haploid, 17 20 survivors in the induced mph1 sample remained haploid, compared to 10 the MPS1 sample. These data suggest that mph1 rescued the SPB duplication defect in S. cerevisiae because if only the checkpoint defect was rescued, mps1-1 cells with a monopolar spindle would be expected to arrest with a 2C DNA peak upon shift to the nonpermissive temperature. We cannot distinguish between rescue of the SPB defect alone, and rescue of both the SPB and checkpoint defects in this experiment. Therefore, we tested the ability of mph1 to complement a second class of mutation in MPS1 distinct from the conditional lethal mutation, mps1-1. Mps1-pac8 was isolated as a mutation that is lethal in combination with a deletion of the CIN8 gene, which encodes a kinesin-related motor involved in spindle pole separation during mitosis, but has no phenotype on its own Geiser et al., 1997 ; . The mps1-pac8 cin8 null mutant strain is kept alive by the expression ofCIN8 from a plasmid that can be counterselected by growth on cyclohexamide containing plates, meaning that the non-transformed strain is inviable on cyclohexamide plates Fig. 5C ; . Transformation of the strain with a plasmid containing MPS1 rescues viability, while transformation with a plasmid containing mph1 results in a partial rescue of viability Fig. 5C ; . Taken together, these results showing partial complementation by the mph1 gene of two different mutations in MPS1, in conjunction with the above experiments testing MPS1 function in S. pombe, demonstrate that mph1 and Mps1p show functional as well as sequence conservation. dph1 is structurally and functionally relataed to S. cerevisiae Dsk2p Partial sequence analysis of the dph1 cDNA showed that the corresponding genomic DNA sequence had been determined by the S. pombe genome project SWISS-PROT accession number Q10169 ; .
