Cellular signal transduction pathways are characterized by their high complexity. Mathematical models describing these processes might be of great help to gain qualitative and, most importantly, quantitative knowledge about such complex systems. Detailed mathematical description of such systems leads to very large systems of ordinary differential equations. The situation becomes even more complicated when we consider two or more pathways simultaneously. Therefore, it becomes necessary to simplify the models, but in such a way that the reduced models still capture essential dynamics. In addition, due to the sparseness of appropriate experimental data, a subset of the model parameters needs to be determined using mathematical optimization. We are interested to find the best possible way to characterize the accuracy observability of the model parameters. Models of signal transduction will be considered in the context of biomarker identification for drug therapy. In particular, model predictions will be used to contribute to better understanding of molecular mechanisms of cancer and linezolid.

Composite materials have a higher specific strength and lower weight than their metallic counterparts and are finding increased usage in aircraft structures. Composite structures can be significantly lighter, which translates to lower fuel consumption and operating costs. In the past, composite materials were confined to secondary structures in aircraft, but composites are now finding greater usage in primary structures, such as the wing box in the Airbus A380. Also, the new Boeing 7E7 aims to utilise composites in 50% of its airframe weight. However, the design of composite components is much more difficult than that of metals due to the anisotropic mechanical response of composite shells, complex failure processes and a large variety of possible materials, geometries and lay-ups. As a result, more advanced design and failure prediction techniques are required than those traditionally used for metals. To reduce development and certification costs for composite structures, efficient computational methods are required by the aircraft industry to predict structural integrity and failure initiation and progression to final fracture when subjected to complex loading. Final fracture mechanisms normally involve delamination at some stage in the failure process, so it is critical that a computational model should have the capabilities to predict this behaviour. A numerical model has already been developed to capture a number of intralaminar failure mechanisms through progressive damage modelling. This is implemented using a user-defined subroutine and ABAQUS finite element FE ; software. A natural progression to this work is to incorporate interlaminar failure as a failure mode in these models to fully capture the composite failure mechanisms. Delamination initiation and progression criteria have been assessed from literature and implemented in a progressive damage model, using methods that are practical both in terms of model definition and computational expense. The laminate is modelled as separate layers connected by a user-defined interface, with each layer representing a single ply of the laminate. By assigning damage and fracture properties to the interfaces, the layers can debond and introduce a delamination in the structure. Thus, both damage and fracture mechanics are incorporated to suitably capture the behaviour of the delamination front. For validation purposes, results obtained using this model are compared with the work of other authors, including both experimental and numerical results for single-mode and multimode delamination testing. The interface model is also employed in a practical application involving pinloading of a composite joint. It is used to accurately predict delamination initiation for a number of stacking sequences.
